On the hiatus between seasons 2 and 3, The CW cast Odette Annable as Reign, and though the actress made her debut as the seemingly human Samantha Arias in the Supergirl season 3 premiere, the creators behind the series have made no attempts to hide that the character will evolve into a major antagonist this season. But beyond the setup, there could be some things that Supergirl takes from the comics in order to make Reign a particularly memorable villain.
The CW's superhero drama has revealed a first look at single mom Samantha's transformation into the ruthless Worldkiller she's destined to become, and based on her appearance alone it's clear that the Supergirl showrunners have chose to take the mythology in a different direction than what comic book fans may have been expecting. In the comics, Reign was taken by Kryptonian scientists as a baby and experimented on to become a new Worldkiller - alongside four other babies who also became Worldkillers.
According to the DC Comics description, Reign was one of five Worldkillers created by Zor-El, who later came to regret his part in the creation of those blood-thirsty, super-powerful monsters. Now, we have our first look at Reign on Supergirl.

Instead of the monstrous-looking biological weapon ripped from the pages of the comics, Annable's Reign is much more humanized. "And that will place some of our characters at odds with each other". But whether Reign is forced to leave the planet or chooses to, it's clear that in the DC comics version of events, Reign is totally unsuccessful when it comes to fighting Supergirl, and is roundly defeated in just a few issues' time - which may foreshadow her probable defeat by Supergirl in the CW world, too.
